2× YT is richer than LB, but very similar in its composition. It is especially useful for culturing phage-infected bacteria.
To prepare L 2× YT Broth

- Prepare 0.8 L of H2O in a suitable container.
- Add 16 g of tryptone to the solution.
- Add 10 g of yeast extract to the solution.
- Add 5 g of Sodium chloride to the solution.
- Adjust pH with NaOH sol. to 7.0.
- Add H2O until the volume is 1 L.
- Autoclave for 15 min on liquid cycle.
